OBJECTIVES

- Master at a theoretical and practical level the NCL and SEA ligations and the desulfurization reactions
- Be able to adapt the protocols to the targeted objectives

AUDIENCE

PRE-REQUIREMENT

Master the basic notions of synthetic organic chemistry (bachelor's degree). Master the theory and practice of the solid phase peptide synthesis (see for example the internship n° 17174 within this catalogue)

TRAINING PROGRAMME

Theory (1 day)
- Native chemical ligation (NCL)
- SEA ligation
- SeEA ligation (selenium version of SEA ligation)
- Application of NCL and SEA ligation, including the latent properties of the SEA group, to the total synthesis of proteins, cyclic peptides, branched peptides and cyclic and branched peptides.
- Desulfurization/deselenization reactions

Practice (3 days)
- Synthesis, cleavage, deprotection and purification of SEA peptides (includes the monitoring of the rearrangement of the SEA group in acidic media by LC-MS)
- Synthesis of peptide thioesters by SEA/thiol exchange
- NCL
- SEA ligation
- Sequential NCL/SEA ligation for the synthesis of large peptides by a one-pot three peptide segments assembly approach
- Desulfurization allowing to convert a Cys residue into Ala
